Overview:

SignalChem Diagnositcs Universal One-Step Cloning Kit is an advanced homologous recombination cloning tool. Its optimized 2X premix combines a recombinant enzyme, buffer, and a recombinant enhancer that significantly boosts cloning efficiency. This kit enables direct cloning of PCR products into any vector at any desired site. It is compatible with unpurified PCR products, directly recovered PCR fragments, and low-concentration gel-extracted DNA. This system efficiently reassembles fragments with homologous arms with 30–70% GC content. The vector is fully linearized, and homologous sequences (15–25 bp) at the ends of the vector are incorporated into the 5' ends of the forward and reverse PCR primers, ensuring that the 5' and 3' ends of the PCR products match the linearized vector. With the action of the recombinant enzyme, the cloning reaction can be completed in as little as 5 minutes at 50°C, achieving a positive cloning rate of over 95%.
