Overview:

SignalChem Diagnostics DNase I (Deoxyribonuclease I), GMP-Grade (2 U/μL) is a high-purity, recombinant enzyme designed to degrade and remove DNA contaminants from RNA and protein samples, ensuring optimal purity for downstream applications. Provided in the aqueous form, it is an endonuclease that digests both single-stranded and double-stranded DNA, breaking them down into single deoxynucleotides or short single- or double-stranded oligodeoxynucleotides. DNase I hydrolyzes phosphodiester bonds, generating monodeoxynucleotides and oligodeoxynucleotides with 5'-phosphate and 3'-OH groups. The smallest digestion product is typically a poly-tetranucleotide.

DNase I can act on various DNA forms, including single-stranded DNA, double-stranded DNA, and chromatin, though its cutting efficiency in chromatin is influenced by histones. It functions optimally at pH 7-8 and requires Ca²⁺ for activity. The enzyme can be activated by divalent metal ions such as Co²⁺, Mn²⁺, and Zn²⁺. 5 mM Ca²⁺ can protect the enzyme from hydrolysis.

In the presence of Mg²⁺, DNase I cleaves DNA randomly at any site on either strand. However, in the presence of Mn²⁺, it cuts both strands at nearly the same location, producing either blunt ends or sticky ends with 1-2 nucleotide overhangs.

DNase I is commonly used to remove template DNA after in vitro transcription and ensure DNA-free RNA for RT-PCR and RT-qPCR. It is also used in DNA labeling via nick translation in combination with DNA polymerase I, as well as in DNA fragmentation for library construction.
