Overview:

This qPCR assay is specifically designed for the rapid and reliable detection of Hop Latent Viroid (HLVd) in Cannabis sativa plant tissue. Utilizing a duplex reaction format, the assay simultaneously amplifies a conserved region of the HLVd genome alongside a Cannabis-specific housekeeping gene, which serves as an internal control. This built-in control verifies the quality of the extracted RNA and detects potential PCR inhibitors that could lead to false-negative results, ensuring high confidence in assay performance. The assay is compatible with one-step RT-qPCR workflows and is optimized for use with high-quality RNA extracted from leaf tissue. Validation was performed using a diverse set of marijuana leaf samples exhibiting a wide range of HLVd titers, confirming the assay's sensitivity and reliability across different infection levels.
