Unit Definition: One unit (U) of the enzyme is defined as the amount that catalyzes the hydrolysis of pyrophosphate (PPi) to release 1 μmol of orthophosphate (Pi) per minute. The standard reaction is carried out in a 500 μL system containing 100 mM Tris-HCl (pH 7.2), 2 mM MgCl₂, and 2 mM PPi, incubated at 25°C for 10 minutes.

2. The optimal reaction temperature is 25°C, though the enzyme remains active between 16°C and 37°C. Heating to 65°C for 10 minutes will inactivate the enzyme.
